Analysis of Lipid Metabolism in Brain of p25 Transgenic Mouse- A Model of Alzheimer’s Disease

Alzheimer’s disease (AD) is an age-associated disorder that leads to neurocognitive impairment. Its main pathological hallmarks include amyloid beta (Aβ) plaques and hyperphosphorylated tau forming intracellular neurofibrillary tangles (NFT). Hyperactivation of a tau-phosphorylating protein, cdk5 by its activator p25 has been shown to be associated with AD. Cdk5 is believed to be playing an important role in regulating lipid metabolism. Changes in lipid metabolism are also increasingly being implicated with neurodegeneration. We analysed whether hyperactivation of cdk5 by p25 overexpression leads to changes in lipid metabolism using mass spectrometry and gene array technologies. Our results showed major changes in cholesterol metabolism and transport, levels of lysophospholipids and PC metabolism. These data will provide clues into the neurodegenerative process with regards to dysfunctional lipid metabolism.
